چکیده

Fluoride which is widespread in our environment and food due to its geological origin industrial pollution has been identified as a developmental neurotoxicant. Gut-brain axis provides new insight into brain-derived injury. We previously found the psychoactive effects of probiotic strain, Lactobacillus johnsonii BS15 against fluoride-induced memory dysfunction mice by modulating gut-brain axis. In this study, we aimed detect link between reconstruction gut microbiota through alleviate impairment. also added an hour water avoidance stress (WAS) before behavioral tests sampling, aiming demonstrate preventive on impairment after psychological stress. Mice were given fluoridated drinking (sodium fluoride 100 ppm, corresponding 37.8 ± 2.4 ppm F¯) for 70 days administered with PBS or 28 prior throughout day exposure sodium fluoride. Results showed that increases hyperactivity hypothalamic-pituitary-adrenal (HPA) reduces exploration ratio novel object recognition (NOR) test spontaneous during T-maze following WAS, significantly improved probiotic. 16S rRNA sequencing significant separation ileal fluoride-treated control mice. was main targeting bacteria reduced reconstructed fluoride-post increased relative abundance Lactobacillus. D-lactate content diamine oxidase (DAO) activity, two biomarkers permeability serum probiotic-inoculated ZO-1, intestinal tight junction protein mRNA, levels treatment. Moreover, hippocampus essential learning memory, down-regulated mRNA level both myelin-associated glycoprotein (MAG), neurotrophic factor (BDNF), including improvement cAMP response element-binding (CREB) fluoride-exposed WAS. Via spearman correlation analysis, displayed positive associations tests, nerve development related factors, proteins negative association TNF-? hippocampus, highlighting regulatory potential barrier. These results suggested addition, there may be some correlations microbiota. reads have uploaded NCBI. Cognitive impairment by antibiotic-induced gut dysbiosis: Analysis of gut microbiota-brain communication

Emerging evidence indicates that disruption of the gut microbial community (dysbiosis) impairs mental health. Germ-free mice and antibiotic-induced gut dysbiosis are two approaches to establish causality in gut microbiota-brain relationships. However, both models have limitations, as germ-free mice display alterations in blood-brain barrier and brain ultrastructure and antibiotics may act direc...

Regulation of gut luminal serotonin by commensal microbiota in mice

Gut lumen serotonin (5-hydroxytryptamine: 5-HT) contributes to several gastrointestinal functions such as peristaltic reflexes. 5-HT is released from enterochromaffin (EC) cells in response to a number of stimuli, including signals from the gut microbiota. However, the specific mechanism by which the gut microbiota regulates 5-HT levels in the gut lumen has not yet been clarified. Our previous ...

Kisspeptin-13 ameliorates memory impairment induced by streptozotocin in male rats via cholinergic system

Introduction: Kisspeptin-13 (KP-13) is a novel endogenous factor, increases synaptic transmission and is involved in several behavioral functions such as anxiety, locomotion, epilepsy and avoidance learning. However, the role of this peptide on cognition has not been well clarified yet. Here we studied the effect of kisspeptin-13 pretreatment on spatial learning and also interaction with cholin...

The effect of intracerebroventricular administration of insulin on memory impairment-induced by scopolamine in male rats

Introduction: Cholinergic neuronal deficiency is one of the main causes of Alzheimer's pathology, which leads to learning and memory impairment. Scopolamine is a muscarinic cholinergic antagonist commonly used to induce Alzheimer's disease (AD). Insulin also regulates learning and memory function. Thus, the aim of this study was to determine the effect of central administration of insulin on pa...
